Abstract: This movie is a time lapse recording of a human MRC5 nucleus labelled with cerulean-tagged PCNA. The cells are grown on glass bottomed dishes in a CO2 and humidity controlled chamber on an inverted Leica SP5 confocal microscope. Midplane sections were acquired every 6 minutes and the sequential images were registered using Image J software to correct for the cells movement over time. Replication factories are not visible at the start of the movie as the cell is in G1 phase, DNA replication has not begun. Replication factories form as the cell starts synthesising DNA to copy its genome. These are seen as bright dots or foci containing high concentration of the fluorescently tagged protein cerulean-PCNA. PCNA is a protein which becomes concentrated in these factories as it is an essential factor required for many of the steps of DNA replication. The entire S phase takes approximately 6 hours, after which the factories disappear and the PCNA once again becomes diffuse throughout the nucleus as the cells enter G2 phase.
